Instrument and equipment

Experimental materials
Sucrose, agar, boric acid, hydrochloric acid, distilled water for chemical analysis.
Culture medium preparation
With 50ml of distilled water by adding 0.5 grams of agar 5g sucrose dissolved after heating, adding sucrose to be completely dissolved, adding 0.01% boric acid with hydrochloric acid pH modulation of 5.5-6.0 coated on glass slides.
The experiment steps
1, to medium temperature culture on a glass slide to room temperature after the trace pollen sprinkled gently on culture medium.
2, to the culture dish into the absorbent paper and pour distilled water will be sprinkled with moisture, pollen slides are placed in a Petri dish, with the lid, the Petri dish placed in constant temperature incubator, 25 C culture 4h.
3. The number of pollen cells and the number of germinated pollen cells were counted by microscope.
Germination percentage (%) = (Q/W) *100
Formula
